What is Membrane Filter?

How to Store the Chromatography Column?

1.Prevent the generation of microorganisms from the buffer solution and the mobile phase of the aqueous solution, make sure that the mobile phase is used in a small amount, or add 200 ppm of sodium azide to the mobile phase of the water to inhibit bacterial growth (beware of its toxicity and potential explosiveness); or in the mobile phase Adding 20% of the organic modifier can also inhibit the growth of microorganisms. The organic modifier also helps to degas the mobile phase.

2.Store the column as much as possible in 100% organic solvent (Beiqing best) and avoid storing in buffer solution.

3.After the buffer solution was applied to the column, 15 to 20 column volumes of the same kind of water-organic solvent mobile phase containing no buffer were used to wash the column and then stored in a 100% organic solvent.

4.Avoid flushing tightly bonded C18 columns with purified water.

5.Twist the ends of the column with a plug to prevent the column bed packing from cracking.

Applications of Vacuum Pumps

In the field of oil regeneration and rerefining, vacuum pumps create a low vacuum for oil dehydration and a high vacuum for oil purification.[12] Especially in the field of transformer maintenance, vacuum pumps play an essential role in transformer oil purification plants which are used to extend the lifetime of transformers in the field.[13]


Vacuum may be used to power, or provide assistance to mechanical devices. In hybrid and diesel engine motor vehicles, a pump fitted on the engine (usually on the camshaft) is used to produce vacuum. In petrol engines, instead, vacuum is typically obtained as a side-effect of the operation of the engine and the flow restriction created by the throttle plate, but may be also supplemented by an electrically operated vacuum pump to boost braking assistance or improve fuel consumption. This vacuum may then be used to power the following motor vehicle components:[14] vacuum servo booster for the hydraulic brakes, motors that move dampers in the ventilation system, throttle driver in the cruise control servomechanism, door locks or trunk releases.


In an aircraft, the vacuum source is often used to power gyroscopes in the various flight instruments. To prevent the complete loss of instrumentation in the event of an electrical failure, the instrument panel is deliberately designed with certain instruments powered by electricity and other instruments powered by the vacuum source.

Prepacking the Flash Column

  • Remove the end fittings ;
  •  Eliminates the thermal effect from solvent adsorption on a dry cartridge;
  • Ensures pre-equilibrium between the solvent and the silica .
  • Entrapped air ,the thermal effect and lack of equilibrium can cause separation, purity and recovery  problems ,especially with early eluting compounds .
  • In general the recommended pre-equilibration volume is 3-5 column volumes prior to sample loading. If using gradient elution ,pre-equilibrate with the starting solvent mixture.

  • Liquid loading: Each column is supplied with a built-in inserter which allows liquid injection via a syringe.
  • Dry loading: If internal dry loading is to be used,this inserter must be removed.

(1) Packing (fixed phase)

Selecting the appropriate SPE column filler is the prerequisite for the ideal result.

(2) The strength of the solvent

1. With positive phase fixation, the strength of solvent increases with its polarity

2. With the reverse stationary phase, the strength of solvent increases with the weakening of the machine

(3) PH value

Ion exchange fixed phase, analyte and interference matter have different PKa. By adjusting the PH, the stationary phase can be charged with opposite charge and the interfering material has no charge. Or, in turn, the stationary phase is charged with the opposite charge, while the analyte does not carry the charge, thus achieving the purpose of separation and purification

Problem When Use HPLC

(1) Mobile phase: must be used after chromatography through the filter after use, and the mobile phase ratio can not be completely in accordance with the standard.


(2) Abnormal column pressure: the bubble inside the column, with the syringe to the bubble out, the column pressure back to normal. Column height: internal obstruction problems occur.


(3) Accuracy of external standard method: the accuracy of the external standard method related to the determination of the results, so the injection must be accurate results, the standard peak area RSD value should be less than 2.0. The general injector should not have bubbles present when extracting samples.


(4) When the test sample or standard peak pinning phenomenon, should check whether the phenomenon of collapse of the column occurred, if any, then re-column or change the new pillars.
